This management includes cordoning off the area and ensuring that the appropriate specialists, such as forensic scientists or pathologists, are called. In addition, a log book must be kept at the crime scene which indicates the persons who have entered and left the scene. This helps to differentiate between traces left behind by investigators or other persons entering the scene and evidence left behind as a result of the crime itself. All evidence collected must be properly labelled, documented and recorded and photos or sketches of the crime scene should be made. Documentation of evidence is essential as it is useless to analyze a piece of evidence and then not be able to identify its origin. The photos or sketches are useful as they can be used for reference after the crime scene has been cleared. The chain of custody begins at the crime scene at the point of collection. Physical evidence is any object which establishes that a crime has been committed or which links the crime to a victim or perpetrator. There are several types of crime scenes and correspondingly several types of physical evidence. The types of crimes include sexual assault, murder, drug raids, arson, fraud and robbery. The types of evidence collected in relation to these crimes include hair, semen, blood, fibres, fingerprints, narcotics, empty gasoline bottles, documents and tools.

Bloodstain pattern analysis refers to the examination of shapes, locations and patterns of bloodstains in an attempt to provide an explanation of the physical events which caused the stains. This analysis is based on the premise that the patterns are characteristic of the forces that created them. There are several classifications of bloodstain patterns. This force may be either internal or external and the pattern seen depends on the amount of force which was exerted. Medium velocity forces of 5 to 25 feet/sec usually result in preponderant stain sizes of 1 – 4 mm, whereas high velocity forces such as approximately 100 feet/sec would result in preponderant stain size usually of less than or equal to 1 mm. High velocity forces occur for example in the case of gunshots. Altered patterns may also be seen which are physically or physiologically changed. These changes may arise from blood clots, diluted stains (such as if someone was trying to clean up the blood), insect activity or as a result of obstruction by an object.

These factors are important in order to maintain the integrity and value of the evidence collected. These factors are:
Protection = avoid contamination of the evidence
Procurement = ensure that the collected evidence coincides with the documentation

Blood:
Preliminary examination for blood first involves screening using the Kastle Myer test. This is based on the property of haemoglobin which enables it to accelerate the oxidation of phenolphthalein, in a peroxidise-like manner, such that there is a colour change from colourless to pink. In cases where the Kastle Myer test gives a negative result, but the presence of blood is thought to be likely the luminol test is employed. This test is not the first choice test since it degrades the DNA present, but it is very sensitive and can detect tiny traces of blood up to 300,000 times diluted. The luminol reacts with haemoglobin to produce molecules which emit light.
After a positive preliminary test confirmation testing must be done. This involves determining the species of origin of the blood in question, which can be done using the precipitin test. The precipitin test involves an immuno-electrophoresis reaction using an antigen-antibody reaction. The antibody may be an anti-human serum or may be for other animals which could possibly have been present. In a positive reaction a precipitate is produced.

Weapons:
Weapons found at a crime scene must be handled in the same manner as all forms of evidence collected. Some weapons may have biological evidence such as bloodstains which can be analyzed while others are sent straight to the Ballistics Department. The test is based on sodium alpha-napthyl phosphate and fast blue dye, which turns purple in the presence of acid phosphatase.

Swab and Smear:
Swabs and smears are used to facilitate microscopic examination, often especially in cases of sexual assault. In most cases the doctor who examines the victim takes swabs of specific areas including vaginal, anal, penile and oral/buccal, depending on the case. For some cases the oral swab can be used for comparison. Swabs are usually examined for the presence of spermatozoa. Fibre samples are analyzed to see if they are natural or synthetic (and the class of synthetic fibre), used for colour comparison and used for texture comparison with statements given.

DNA:
DNA contains all the genetic information of the cell and is made up of genes and alleles, which are variations of genes. The DNA is tightly coiled into chromosomes which vary in number from one organism to another. DNA can be used for several applications including forensic investigations, paternity testing, identification of missing persons or catastrophe victims and lineage studies. Humans have 22 pairs of autosomal chromosomes and one pair of sex chromosomes. DNA also contains short tandem repeats (STR) which vary in length and number from one person to another. These STR markers are therefore useful in DNA analysis and currently eight STR markers, each with a variety of sizes or alleles, are being used for analysis. For example, TPOX is an STR found on chromosome 5 and its repeat unit is AATG and this unit can be repeated between 6-13 times. The STR may be repeated six times o one homologous chromosome and eight times on the (one from each parent) but it is still the same gene on both chromosomes, only the STR size is different. In this example the genotype for TPOX would be “8,6″. When the DNA is separated by gel electrophoresis the larger sizes move slower on the gel. Sources of biological evidence include blood, semen, urine, hair, fingernails, tissue, saliva, teeth and organs. The processes involved in DNA analysis are extraction, quantification, PCR and STR typing. DNA extraction can be done using several methods.

DNA Extraction:
Firstly, the Chelex (inorganic) method can be used. It involves the use of PBS (phosphate buffer solution) to free the cells from the sample. The sample is then boiled in a solution containing chelating resin beads that bind metal ions such as magnesium which causes the cells to lyse and release the DNA. This is not a very efficient method as it doesn’t produce much DNA. The second method is the organic method which uses phenol chloroform iso-amyl (PCI). PCI precipitates proteins, fats and extraneous materials thus leaving only the DNA in solution. Alcohol is then used to clean and precipitate the DNA, thereby increasing its purity. Thirdly, the DNA IQ (Promega) method can be used. A lysis buffer containing a salt (guanidine thiocyanate – GTC) is used which causes the DNA to stick to the paramagnetic resin. Following this a wash is done with low salt or alcohol based wash buffer and the DNA is isolated. Finally, the Maxwell 16 method can be used which uses pre-dispensed reagent cartridges and has moderate throughput. The inorganic and organic methods are older and the DNA IQ and Maxwell 16 methods have now been developed for several reasons. The older methods involved procedures such as soaking samples, which results in inefficient DNA recovery and may cause significant loss of DNA. The older methods also can introduce amplification inhibitors and in addition the chemical PCI is hazardous. The newer methods allow for DNA extraction without these problems.

DNA Quantification:
DNA quantification is used to determine the amount of amplifiable DNA. 0.1 to 0.3 ng of DNA is required to get accurate results in the analysis. A kit called the Plexor HY from Promega is used with Corbett Rotor Gene 6000 Real-Time PCR. During the PCR, as DNA is amplified the fluorescence decreases. The reporter used is iso-dC and the free nucleotide dabcyl-iso-dGTP binds to the reporter at the end of amplification. This is known as fluorescence quenching.

PCR:
PCR is used to copy target STRs through thermal cycling. The components used for PCR are nuclease-free water, STR buffer containing dNTPs and Mg2+, primers and Taq DNA polymerase. The primers and Taq DNA polymerase enable stringent copying of only STR sequences. The primers are tagged and have an affinity for the markers. They are tagged by two fluorophores, namely tetramethylrhodamine (TMR) which is red and fluoroscein (FL) which is green. Four of the eight STRs are complementary to the primers with TMR, while the other four are complementary to the primers with FL. These tags aid in band visualization after electrophoresis.

STR Typing:
STR typing is mainly done using two methods. Firstly, polyacrylamide gel electrophoresis method can be used. In this method the sample must be denatured and mixed with a loading dye (often bromophenol blue) which keeps the sample in the well by making it heavier. The samples can then be loaded and the electrophoresis is carried out followed by scanning and examination of the gel. In this method another fluorophore called carboxy -X- rhodamine (CXR) is used to tag the internal lane standard and is added to each well. The FMBIO machine is then used which recognizes the different wavelengths of the fluorophore tags and converts this data into digital data and visualizes the colours. It is important to use these fluorophores since some of the STRs will be the same size and therefore will move the same distance on the gel. Without the colour difference it would not be possible to identify them as separate STRs. The four which are tagged green are of varying sizes and so they will not overlap with each other. The same is true for the four which are tagged red. After the visualization is done the genotype must be derived from the images. This is done by identifying the two alleles of each of the eight STRs and the resulting data would be as follows, for example for CSF – 8,6.
The second method used is the ABI Genetic Analyzer 3100. In this method the amplified DNA fragments are loaded into the machine which has 16 silica capillaries which enable the separation of the fragments by electrophoresis.

Y-STRs
The Y chromosome is found only in males and is therefore passed on from father to son. Due to this fact the Y chromosome can offer particular information which is useful to several applications including genetic genealogy studies. Analysis of Y-STRs is especially useful in sexual assault cases which do not have clear results. Since only the Y-STRs will be tested for, the female victim’s profile will not interfere with the analysis. Y-STRs are smaller than the other STRs and will remain in tact even in highly degraded samples and 9 core Y-STRs are used in a reference database. The advantages of using these are that it makes it easier to exclude suspects and easier mixture analysis. However, the disadvantage to using this method is that it is not easy to include someone since the Y chromosome is the same in each generation.

Science Fiction Classics

I love to read science fiction.  And I was thinking the other day about a question someone asked me – what would you recommend for someone to start out reading science fiction?  What are the classics that define the field?  Could I choose 20 books to start a science fiction library with?

Here are my picks.

The Science Fiction Hall of Fame Volume 1

I start with this one, a short story collection, because of what it contains.  Classics.  In this volume are stories that led to movies, like Charly, based on the short story Flowers For Algernon.  Or It’s A Good Life that appeared as a Twilight Zone episode.  This volume includes such classics as Nightfall, The Cold Equations, and many more.

Pellucidar

By Edgar Rice Burroughs, the author who also created Tarzan.  Pellucidar is the world found at the center of the Earth.  Join David Innes as he travels across the face of Pellucidar meeting and sometimes battling savage tribesman, or Mahars – intelligent reptiles who feast on human flesh.  Read the whole series.

Lensman

Galactic society is held together by a group of interstellar law-beings.  Lensman.  Given a device by an ancient race of super-intelligences these men, women, and beings, fight evil and try to create galactic peace.  Written by E.E. “Doc” Smith, the Lensman series is truly classic space opera.

Earth Abides

The apocalyptic novel following the events of the last American and how he teaches the next generation to survive in this frightening new devastated world.  Written by George Stewart, I think this easily outclasses I Am Legend.

Dune

You’ve probably seen the movie, or the mini-series, why not read the book?  A classic of scope and character.  Follow the death and birth of a dynasty through the lives of the Atreides family, and the fulfillment of prophecy.  Written by Frank Herbert, it, and the other books in the series, deserve a place among the classics.

Foundation

Hari Seldon had a dream.  That the galactic dark ages that were coming would be shortened.  His goal, to set up a Foundation to save the accumulated information of the galaxy in an “Encyclopedia Galactica”.  But was that his real purpose, and what were the rumors about a Second Foundation located at the end of the universe?  Written by the Grand Master of science fiction himself Isaac Asimov.

The Stainless Steel Rat

Harry Harrison brings us the story of Slippery Jim DiGriz, also known as the Stainless Steel Rat.  Take James Bond, Matt Helm, and Derek Flint,  roll them into one man, make him a thief, and put him on the Galactic scene and that is Slippery Jim.  Out-conned into working for the government.

Dorsai!

Gordon Dickson brings us the tales of a race of people bred solely to be the galaxy’s warriors.  Mercenaries for hire.  Bred for strength, intelligence, and trained in the arts of war.  Be sure to read the entire Childe Cycle.

Ringworld

Larry Niven brings us a tale of the discovery of the millennium for the galactic community.  An actual Dyson Ring.  A ring millions of miles long stretched in orbit around a sun inhabited by many different and diverse peoples and species.  Join Louis Wu as he and his team explore this strange new world and determine if the makers are still around.

City

Another master of science fiction was Clifford Simak.  In City, we have a collection of his short stories which follow the exodus of humanity from Earth, and what remains.

Gateway

Mankind went out to the stars and found the Heechee.  Well, they found the Heechee’s technology, and their ships, which all seem to have pre-programmed destinations.  The corporation needs people to get in these ships, push the buttons and explore.  But it’s dangerous as sometimes there are no facilities at the end of some of the routes.  Some places have gone nova.  Frederick Pohl brings us this story about the people who bravely went into the unknown, and hints about what may have happened to the Heechee.

To Cage A Man

F.M. Busby tells us what has happened to all those people who have been abducted.  The Demu have taken them.  But for what sinister purpose?  Until they day they take the man who says – I will not be caged.  This is actually a trilogy with a far-reaching story.  It’s a very good read.

To Your Scattered Bodies Go

Richard Burton died.  Then he woke up and he was naked, next to a river, along with a whole of other naked people.  Who had also been dead.  Determined to find out what is going on and to find the masters of the Riverworld as it comes to be called, Philip Jose Farmer has crafted a very interesting story, that continues in several further books.  Another classic.

Nine Princes in Amber

Have you ever felt that you were living a dream?  That perhaps there was another deeper reality?  Perhaps many deeper and different realities?  Roger Zelazny brings us the story of one man, who awaken in the hospital, with amnesia.  Only to be confronted by someone who tells him he is a prince and must return to Amber, the final reality from whence all other realities derive, and that he has the power to walk through these realities.  He must return because the throne is empty and he is one of nine who might claim the throne.

Childhood’s End

The aliens arrived on earth and to everyone’s surprise, they looked like the demons of old.  But they work for God, or some super alien being, who has determined it is time for the next stage in humanities evolution.  But why do only the children seem to be affected.  Join Arthur C. Clarke with this tale of life, love, and loss, and what it means to be human.

Midnight At The Well Of Souls

Jack Chalker brings us the story of Nathan Brazil, spaceship pilot, and one of the few who knows the planet where the Well of Souls is supposed to reside.  An artificial planet, that changes all who come to it, sometimes physically, sometimes in unseen ways.  What do the planets rulers have in mind for all these beings they are experimenting on?  And why is Nathan called the Wandering Jew?

Lord Valentine’s Castle

Brought to us by Robert Silverberg, Lord Valentine’s Castle brings us to the world of Majipoor.  It begins with Lord Valentine, waking up and finding that not only has he been mugged, but what was taken was his body as he is now apparently wearing someone else’s.  Follow Valentine as he seeks to regain not only his body but his rightful place on Castlemount, and determine who is behind this.  Could it be the King of Dreams?

Ender’s Game

Orson Scott Card shows us humanity at war, with the bugs.  Such a costly war that we have taken to training children to fight in it.  The best, tested and trained day after day, simulation after simulation, until one, Ender Wiggin, stands out from the crowd.  But when do they go to fight?  And were those really simulations?

Startide Rising

Welcome to David Brin’s universe of Uplift.  That means that every sentient race in the galaxy can trace it’s lineage back through it’s patrons, the races who tinker with their genetics so the become sentient.  And from there all the way back to the mysterious Progenitors, who started the whole process.  Well all of them except it would seem but humans.  The galactic society is outraged.  Worse yet, humanity has uplifted two of it’s own to sentience, dolphins and chimpanzees.  This is the tale of the starship Streaker, a ship crewed by mainly dolphins, who have stumbled across a secret almost any race in the galaxy would kill for – a fleet of derelict space ships, 50,000 of them.

On Basilisk Station

David Weber re-invents space opera with the Honor Harrington saga.  Meet Honor, a young woman who is a whiz at commanding a spaceship and getting it out of the toughest situations, all the while avoiding the enemies in her own fleet.

And so we have come full circle, going from space opera back to space opera.  I hope if you haven’t read any of these you will give them a try.  At the very least to understand where the genre has come from.  And if you feel I have one out of place, or missed any, let me know.

The Role of Sciences in Civilization

The role of sciences in civilization is very notable nowadays. Without science, it would be difficult for men to experience life to the best that they could do. Besides, the achievement of a more civilized society depends upon the things that are discovered through science such as computers, televisions, food processors, cameras, electrical paraphernalia, medicines, and many other necessities in life. In this paper, the discussion is focused on the comparison of the work of Edward Jenner’s book entitled Vaccination Against Smallpox with Michael Pollan’s book entitled The Botany of Desire in relation to their views on the role of sciences in civilization.

The two writers has different views about the role of sciences in the civilization of mankind. For Jenner, he was more concerned on the cure of smallpox than any other field of discovery. So many years ago, smallpox has been considered as a deadly disease that kills mostly infants and even children. The government is trying to look for a better cure for smallpox which is considered equal to cancer diseases prior to the discovery of vaccination. Jenner believes that there is no other way to save mankind but to conduct series of clinical and research studies to find cure for deadly diseases. The disease coined as smallpox caught the attention of Jenner and when he finally discovered vaccination as a cure, the World Health Assembly officially declared that the people no longer have to worry on smallpox diseases. Simply put, Jenner used his views regarding innovative medicines to cure smallpox as part of scientific development. As a result, human civilization was made easier and more generations were benefited of the discovery and views of Edward Jenner.

On the other hand, Michael Pollan studied and wrote on how plants have manipulated human beings to achieve evolutionary advantage as much as human beings have manipulated plants. Pollan believes that studying the evolution of different botanical creations is one way of achieving civilization in the society. Basically, Pollan explained the influence and the attraction of plants towards mankind in such a way that we depend on their presence. For instance, Pollan wrote that we are inclined to eat apples because of their sweetness, use the addictive aroma of marijuana leaves, and the delicious taste of potato. Pollan also created a unique view on the essential characteristics of plants that made human beings dependent on their cultivation and fruits. Hence, the foods that we eat most likely came from plants and that we also achieve civilization in terms of food technology and processes through the use of plants.

Lastly, both Jenner and Pollan studied science for purposes of solving human dilemmas. The differences of their efforts lies on the subject and the ideas that they presented to the people. On the part of Jenner, his views are more glued to the discovery of medicine for smallpox while Pollan’s views is more focused on the interdependence of plants and mankind.
